Why is drinking wine a sin in some religious sects...?

...when the first miracle of Jesus Christ was to turn water into wine so they could party it up at a wedding?

Yet the Methodists and others ban drinking wine in any amount. They use grape-juice at communion for example. And then the Catholics and their priests, who I've heard follow the same book, drink wine and other alcoholic beverages and apparently have not sinned.
Update 2: So Ruhman you're saying god made a mistake and changed her mind to ban alcohol 700 years later?
