This is a case of 'Dative', Comparative, and Superlative'.

If your sister is the 'most clever' in the family, then you use the superlative 'cleverest'.

If she is being compared to someone else in the family, then you use 'clever'.

    Remember

    Dative, Comparative, & Superlative.

    Good, better, best.

    Light, lighter, lightest.

    Dark,darker, darkest.

    et. seq.,

While "My sister is the cleverest one in our family" is correct, it could be changed to: "My sister is our cleverest family member."

You can use either, but cleverest would be reasonable, since it might reasonably be expected there are more than two in the family.
