Instead of "building blocks" (C, H2O, e.g.), what signs of life could scientists be looking for, what forms of energy?

Scientists already know there is life. Look around you: The Earth is covered with millions of species of life. How many can you name?

Life is easy to find. Just go to your local park, forest or nature reserve. Plenty of life there. Enough to keep anyone busy and fascinated for a lifetime. You can even grow lifeforms in a jar on your window sill. You need an instrument called a microscope to study it.

No idea why you would ask about life on an astronomy forum, where the instrument of choice is a telescope, and no sign of life has ever been found off Earth, despite centuries of observation.
